چکیده

Previous studies have shown that the metabolism of acetone and acetate by the photosynthetic bacterium, Rhodopseudomonas gelatinosa, requires either light or oxygen as a source of energy (1, 2). The studies also showed that the cleavage of acetoacetate to acetat.e did not require an external source of energy. These observations are consistent with the known thermodynamic properties of the reactions which indicate that the cleavage of acetoacetate is an exergonic reaction. In view of current concepts of acetoacetate metabolism (3), it seemed possible that some of the energy liberated by this reaction could be utilized to satisfy part of the energy requirements for the metabolism of acetone and acetate. The coupling of acetone and acetate metabolism with acetoacetate decomposition, anaerobically in the dark, was investigated by means of C14-labeled acetone and acetate. The results indicate that the hydrolysis of acetoacetate can serve as a source of energy for these reactions. The coupling between acetoacetate decomposition and acetate metabolism was much more efficient than the coupling with acetone metabolism.

Engineering of a modular and synthetic phosphoketolase pathway for photosynthetic production of acetone from CO 2 in Synechococcus elongatus PCC 7942 under light and aerobic condition

Capture and conversion of CO2 to valuable chemicals is intended to answer global challenges on environmental issues, climate change and energy security. Engineered cyanobacteria have been enabled to produce industry-relevant chemicals from CO2 . However, the final products from cyanobacteria have often been mixed with fermented metabolites during dark fermentation. In this study, our engineerin...

Fermentative degradation of acetone by an enrichment culture in membrane-separated culture devices and in cell suspensions.

A mixed culture, WoAct, growing on acetone, consisted of two dominant morphotypes: a rod-shaped acetone-fermenting bacterium producing acetate, and an acetate-utilizing Methanosaeta species. Dense cell suspensions, largely free of the aceticlastic methanogen and supplemented with bromoethanesulfonate, were able to degrade acetone and grow in small volumes in membrane-separated culture devices i...

Anaerobic degradation of acetone and higher ketones via carboxylation by newly isolated denitrifying bacteria.

Five strains of Gram-negative denitrifying bacteria that used various ketones as sole carbon and energy sources were isolated from activated sludge from a municipal sewage plant. Three strains are related to the genus Pseudomonas; two non-motile species have not yet been affiliated. All strains grew well with ketones and fatty acids (C2 to C7), but sugars were seldom utilized. The physiology of...

Catabolic and anabolic enzyme activities and energetics of acetone metabolism of the sulfate-reducing bacterium Desulfococcus biacutus.

Acetone degradation by cell suspensions of Desulfococcus biacutus was CO2 dependent, indicating initiation by a carboxylation reaction, while degradation of 3-hydroxybutyrate was not CO2 dependent. Growth on 3-hydroxybutyrate resulted in acetate accumulation in the medium at a ratio of 1 mol of acetate per mol of substrate degraded. In acetone-grown cultures no coenzyme A (CoA) transferase or C...
